We would usually start with three island nights when travelling from elsewhere in Cambodia. That leaves two full days between the arrival and departure journeys. Four nights buys a third full day; two nights can work, but only one day is entirely yours.
This is a recommendation about time, not a minimum imposed by an operator. If you are already near the port, a short stay may make sense. If you are crossing the country for it, the same two nights deserve more scrutiny.

You may enjoy part of the arrival or departure day too. Treat that as extra time, not the foundation of the plan. This table assumes no hotel or island change during the stay.
Your time on the ground
Arrival and departure are not counted as full sightseeing days. These examples allow a day per connection; check the actual schedules and buffers for your trip.
Here is the simplest shape for a three-night island stay followed by an international flight:
The return transfer and overnight base must work for the actual flight. One mainland night is a useful starting buffer, not a promise that disruption cannot affect you. Longer margins may be sensible when conditions or connections are uncertain.
Start with the kind of stay you want: somewhere with a choice of places to eat, or a hotel where you expect to spend most of your time. Then check its exact arrival instructions. A ticket to an island name is not enough to establish how you reach the room.
Ask the hotel for the arrival pier, recommended operator, final transfer and the cost of that transfer. Check whether it works with your chosen sailing. On the return, confirm the pickup arrangement and how far in advance the operator requires you to reconfirm.
Buva Sea states that its boats make several pier stops and that weather can delay services. Its current terms also specify arrival at the designated pier at least 30 minutes before departure. Those are that operator's conditions, not a universal timetable or boarding rule. Read the operator's terms.
With three or four nights, we would first consider staying in one place. Changing islands adds packing, checkout, a connection and another arrival. It can be worthwhile if the second stay offers something you care about; it does not create an extra beach day.

In a short Cambodia trip, an island is often a choice instead of another region. Keep Angkor, the capital and a proper island stay if those matter most; consider leaving the extra countryside base for another visit.
If you are hoping for riverside time and southern outings rather than a beach stay, compare Kampot. It has a different purpose in the trip and should not be sold to yourself as the same experience without the ferry.
